Trial Application and Expected Outcome
  • At least three candidate LAMP primer sets will be assessed using synthetic Legionella DNA controls. The set with the fastest time-to-result and highest sensitivity will be selected. We expect a sensitivity of at least 50 CFU per reaction and time-to-results of 15–35 minutes depending on control DNA quantity.
  • We will evaluate our sample preconcentration method using water spiked with various amounts of Legionella. Preconcentration involves filtering 100–200 mL of sample through a 0.44 micron filter, passing a wash buffer, backflushing the filter with extraction buffer and ensuring extracts yield comparable LAMP outcomes as with DNA controls.
  • LAMP reagents will be integrated into centrifugal microfluidic chips via lyophilization, for improved shelf-stability and user-friendliness; on-chip LAMP performance will be compared against the previous experiments conducted in microtube format. We expect comparable sensitivity and rapidity and can modify chip designs to accommodate greater sample volumes if necessary.
  • We will assess our microfluidic chips with real-world water samples and compare our results against those obtained through HOKLAS accredited laboratories and quantify agreement. Laboratory results will serve as the gold standard. We expect our assay to have similar performance to those which comply with ISO 11731.
  • The shelf-stability of our microfluidic chips will be assessed. LAMP experiments will be conducted using control DNA fragments and real-world water samples and compared to our prior chip and lab results. We expect a shelf-life at room temperature of at least 3 months.
